ALKALINE CELLULASE FREE XYLANASE FROM BACILLUS SP. ASX42: PROPERTIES, PURIFICATION AND ITS EFFECT ON SEED GERMINATION

Bacillus sp. ASX42 isolated from soil samples of Lake Van shores, Turkey, and the strain producing cellulose-free xylanase enzyme with an optimum pH 9.0, at 60ºC. The molecular weight was estimated as 66 kDa on SDS PAGE analysis. Thermal stability detected average 68.7% 4-60ºC 60% 65-95ºC while observed about 90% between 3.5-13.0 for 15 min. As HgCl2 presented a strong inhibitor activity, Cobalt (132%) Manganese (130%) showed stimulatory effect activity. remaining activity found to be 77%, 89%, 92% in presence EDTA, 1,10-Phenanthroline monohydrate β- mercaptoethanol, respectively. In this study, some known purification materials were compared effectiveness. According results, cellulase-free has shown germination Anagyris foedita Ceratonia siliqua seeds. It also produced whitening wastepaper pulp.

Rhizopus stolonifer was cultivated in wheat bran to produce a cellulase-free alkaline xylanase. The purified enzyme obtained after molecular exclusion chromatography in Sephacryl S-200 HR showed optimum temperature as 45o C and hydrolysis pHs optima as pH 6.0 and 9.0. Xylanase presented higher Vmax at pH 9.0 (0.87 μmol/mg protein) than at pH 6.0 and minor Km at pH 6.0 (7.42 mg/mL) than at pH 9.0.
